With more that 25 years of experience in human tissue disaggregation and selection of specific cell types, Evercyte offers services to establish human primary cell cultures from various tissues and organs. The studies are performed with ethical approval from the local ethical committee and prior given written informed consent by the donor.

General information

Tissue, cell type: Evercyte offers the isolation of specific cell types from various tissues and body fluids

Ethical issues: cells are isolated with ethical approval from the responsible IRB and prior given written informed consent
Isolation method:
mechanical and/or enzymatic dissagregation of tissue specific cell-cell and cell-matrix contacts
Cell enrichment:
use of specific cell culture media and coatings, cell sorting using FACS or MACS, cloning
Characterization:
expression of cell type specific markers and function, authentication, freedom from contaminations

Recent example – isolation of amniotic membrane epithelial cells

Amniotic membrane is sepatated from whole placental tissue followed enzymatic tissue digestion. After 7-10 days cultivation in serum-free cell culture medium, small colonies appear showing the typical epithelial morphology. These cells are enriched  and stained for presence of typical marker proteins such as SSEA-4 (red fluorescence, DNA is counterstained with DAPI).

Recent example – isolation of chorionic plate MSCs

Chorionic plate region is prepared from whole placental tissue and cut into small pieces. The resulting fragments are seeded into culture plates for outgrowth of mesenchymal stem cells. These cells are enriched in vitro and show the typical spindle-shaped mesenchymal morphology.

The number of population doublings that can be performed strongly depends on several parameters such as the cell type, the culture conditions as well as the number of cells that can be isolated / enriched from the primary tissue.

Do I have to add antibiotics to the medium?2021-10-08T09:41:21+00:00

We advise to use media without antibiotics for routine cell culture. Please also see “Guidance on Good Cell Culture Practice” by Coecke et al. (2005) https://pubmed.ncbi.nlm.nih.gov/16180980.
